Default Fault code reader operation

Have one of the above on the way. Can these readers be plugged in and then the car driven to "induce" the EML lamp on the dash? And would this then produce the relevant fault code on the reader?
Have a misfire under throttle (not ignition related and not vacuum hoses) and am wondering if maybe a dodgy lambda sensor is to blame (occasionally lights up the EML - briefly). Would these fault code readers be of help in identifying same? (Apologies for my ignorance, should what I'm proposing sound absurd!).
